Where the number sixteen comes from
The 841 mm width of A0 is four times the 210 mm short edge of A4, which is 840. The 1189 mm height is four times the 297 mm long edge, which is 1188. The A series loses a millimetre at each halving, so the numbers are one millimetre out, which never matters in practice: think of it as a 4 by 4 grid. Add the default 5 mm margin and 5 mm flap, though, and it climbs to a 5 by 6 grid of around thirty sheets, which is when switching the output to A3 starts to make sense.
Sixteen sheets need a backing board
Glued edge to edge with nothing behind it, a poster this big ripples under its own weight and the seams start to open while it is on display. Mount the sheets one at a time onto foam board or a large sheet of backing paper to keep it flat. If it is going to be reused at several events, laminate the assembled poster or mount it on self-adhesive fabric poster stock.
The honest cost in time and ink
A consumer inkjet takes one to two minutes per full-colour A4 page in high-quality mode. Thirty pages is close to an hour of printing and most of an ink set. For a single use, tiling wins; for something you will reprint, a print shop is better value. An A0 from a shop typically costs thirty to fifty dollars.

Sixteen in theory, in a 4 by 4 grid, with zero margin and zero overlap. Real printers keep an unprintable border, so the defaults produce nearer thirty. Switching the output sheet to A3 brings it down to roughly eight to twelve.
Is gluing thirty sheets really worth it?
Honestly, it is a chore. Printing on A3 at a copy shop halves the count. But there are situations where making an A0 at home is the only option: a last-minute correction, confidential material you cannot send out, or work at two in the morning.
Will thirty seams ruin the readability?
From two metres or more, almost never. If you are showing dense tables or charts, adjust the source layout so a seam does not land on a row of figures. Raising the overlap to 10 mm widens the shared strip and keeps white gaps from appearing.
